Human Genome Epidemiology (HuGE) Review Contribution of Transforming Growth Factor α Polymorphisms to Nonsyndromic Orofacial Clefts: A HuGE Review and Meta-Analysis

چکیده
We performed ameta-analysis of the association of transforming growth factor α gene (TGFA) polymorphisms with the risk of cleft lip with or without cleft palate (CL/P) or cleft palate (CP). In total, data from 29 studies were pooled for the following 3 polymorphisms: TGFA/TaqI, TGFA/BamHI, and TGFA/RasI in the TGFA gene. A fixed-effects or random-effects model was used to calculate the pooled odds ratios based on the results from the heterogeneity tests. A significantly increasedCL/PorCP riskwas observed in persons carrying aC2 allele at the TaqI polymorphism (odds ratio (OR) = 1.70, 95% confidence interval (CI): 1.41, 2.05) compared with those with a C1 allele (OR= 1.57, 95% CI: 1.23, 2.01). For the TGFA/BamHI polymorphism, carriers of the minor A1 allele had an estimated relative decrease inCL/P risk (OR= 0.44, 95%CI: 0.30, 0.64). These associations remained significantwhenonly high-quality studies were included. However, no significant association was observed between the TGFA/RasI variant and CL/P risk. In summary, this meta-analysis provided a robust estimate of the positive association of the TGFA/TaqI polymorphismwith bothCL/PandCPand suggests that personswith anA1 allelemay have amarkedly decreased riskof CL/P.

Association between the transforming growth factor alpha gene and nonsyndromic oral clefts: a HuGE review.
Transforming growth factor alpha (TGFA) is a well-characterized mammalian growth factor. Since the first report of an association between DNA sequence variants at the TGFA genetic locus and nonsyndromic oral clefts, 47 studies have been carried out, producing conflicting results. In this review, the author synthesizes findings from published reports on the association between the TGFA gene and ...
متن کاملHuman Genome Epidemiology (HuGE) Review Associations Between Inflammatory and Immune Response Genes and Adverse Respiratory Outcomes Following Exposure to Outdoor Air Pollution: A HuGE Systematic Review
Variants of inflammatory and immune response genes have been associated with adverse respiratory outcomes following exposure to air pollution. However, the genes involved and their associations are not well characterized, and there has been no systematic review. Thus, we conducted a review following the guidelines of the Human Genome Epidemiology Network. Six observational studies and 2 interve...

متن کاملBMP7 Gene involved in nonsyndromic orofacial clefts in Western han Chinese
BACKGROUND Nonsyndromic orofacial clefts (NSOCs) are the most common craniofacial birth defects with complex etiology in which multiple genes and environmental exposures are involved. Bone morphogenetic protein 7 (BMP7), as a member of the transforming growth factor-beta (TGF-beta) superfamily, has been shown to play crucial roles in palate and other orofacial ectodermal appendages development ...
متن کاملHuman Genome Epidemiology (HuGE) Review Tumor Necrosis Factor Polymorphisms and Alcoholic Liver Disease: A HuGE Review and Meta-Analysis
The association between alcoholic liver disease (ALD) and tumor necrosis factor-a gene (TNFA) polymorphisms has been analyzed in several studies, but results have been conflicting. The main purpose of this study was to integrate previous findings and explore whether these polymorphisms are associated with susceptibility to ALD. The authors surveyed studies on the relation between TNFA gene poly...
